Validation of mRNA display hits will be conducted using an orthogonal method: ELISA, which offers scalability.

1. Top 90 ~ 900 molecules undergo hit confirmation.

2. DNA synthesis will be carried out for each confirmed hit. Hits will be transcribed and phenotypes formed akin to the process in mRNA display.

3. The binding between targets and hits in each well of the ELISA plate will be assessed.

4. Additionally, the system is adaptable to switch to luminescence signal reading for a broader and more dynamic detection range.
